TY - THES T1 - Nachweis von HERV-K(HML-2) Proteinen und RNAs in der HMM APOBEC3G Granula AU - Block, Adriana AB - Etwa 8% des menschlichen Genoms bestehen aus Sequenzen von humanen endo-genen Retroviren (HERV). Ein Provirus, das für alle viralen Proteine einen vollstän-dig offenen Leserahmen besitzt, ist HERV-K113 aus der HERV-K(HML-2) Familie. HERVs werden in gesunden Zellen epigenetisch kontrolliert, können aber in einigen Tumoren und Autoimmunerkrankungen reaktiviert werden und virale Partikel bilden. In diesem Zusammenhang wird eine kausale Rolle dieser Viren bei der Entstehung und Progression dieser Erkrankungen diskutiert. Bei der Transkription des Provirus im Zellkern werden Volllängen-RNAs gebildet, von denen ein Anteil gespleißt wird. Das akzessorische Protein Rec von HERV-K(HML-2), welches aus einer vollständig gespleißten RNA synthetisiert wird, ist zu-sammen mit dem zellulären Staufen-1 Protein am Transport der nicht bzw. teilweise gespleißten viralen RNA-Transkripte aus dem Zellkern in das Zytoplasma beteiligt. Der Staufen-1/Rec-Ribonukleoproteinkomplex wird im Zellkern gebildet und ins Zy-toplasma zum Ort der Proteinsynthese bzw. Partikelbildung transportiert. In einer zellulären Stresssituation fusioniert dieser Komplex wahrscheinlich mit den APOBEC3G Proteinen und bildet die sogenannte HMM APOBEC3G Granula. In dieser Arbeit wurden zunächst mit APOBEC3G direkt oder indirekt assoziierte Proteine erfolgreich aufgereinigt. HMM APOBEC3G Granula besteht aus einer Viel-zahl von Proteinen und RNAs. Als APOBEC3G assoziierte Proteine wurden G3BP, Staufen-1 und das Rec Protein von HERV-K113 nachgewiesen. Die Co-Aufreinigung von G3BP legt die Vermutung nahe, dass es sich um eine Stressgranula handelt, da G3BP ein Stressgranula assoziiertes-Protein ist. Die Co-Aufreinigung von Rec und Staufen-1 mit APOBEC3G bestätigt zudem kürzlich pu-blizierte subzelluläre Co-Lokalisationsdaten, die alle drei Proteine in der Stressgranula zeigen. Darüber hinaus ist dies eine weitere Evidenz für eine Verbin-dung zwischen dem Rec-asssoziierten RNA-Transport und der HMM APOBEC3G Granula. Diese Hypothese wird durch Experimente zur Detektion vom HERV-K(HML-2) RNA in den APOBEC3G-Präzipitaten in dieser Arbeit erhärtet. Darüber hinaus konnte gezeigt werden, dass durch zusätzliche Transfektion von Rec eine erhöhte Präsenz von HERV-K(HML-2) RNA in der HMM APOBEC3G Granula er-reicht werden kann. Es konnte außerdem zum ersten Male gezeigt werden, dass durch ektopische APOBEC3G Expression die HERV-K113 Partikelproduktion verringert wird. Das könnte ein Hinweis darauf sein, dass der HMM APOBEC3G Restriktionsmechanis-mus auch gegenüber humanen endogenen Retroviren wirksam ist. Durch die Bindung der viralen RNAs in der HMM APOBEC3G Granula steht diese nicht mehr für die Proteinsynthese bzw. Partikelproduktion zur Verfügung. Das führt zu einer Ver-ringerung der gebildeten Viruspartikel und könnte so einen Schutzmechanismus vor Re-Infektion und Re-Integration darstellen. AB - Human endogenous retrovirus sequences (HERV) represent about 8 % of the hu-man genome. The HERV-K113 provirus belongs to the HERV-K(HML-2) family and contains an open reading frame for all viral proteins. HERVs control epigenetically in healthy cells. However, they can reactivate in some tumors and autoimmune dis-eases and can produce viral particle. In this context a causal role of these viruses in the formation and progression of tumors is discussed. The provirus has been transcribed into full-length RNA and a certain percentage of the transcripts can spliced. The accessory protein Rec from HERV-K(HML-2) is syn-thesized from a totally spliced RNA transcript. In conjunction with Staufen-1, Rec is involved in the transport of not or partially spliced RNA transcripts from nucleus to cytoplasm. The Staufen-1/Rec ribonucleoparticle (RNP) formation occurs in the nu-cleus and is transported into the cytoplasm to a place of protein synthesis or particle formation. In a cellular stress situation this complex can be combined with the APOBEC3G protein and forms HMM APOBEC3G granules. First of all, APOBEC3G in conjunction with direct or indirect associated proteins were successfully purified. HMM APOBEC3G granules consist of a multitude of pro-teins or RNAs. G3BP, Staufen-1 and Rec protein from HERV-K113 were detected as APOBEC3G associated proteins. The co-purification of G3BP indicated that the-se are stress granules because G3BP is a stress granule associated protein. The co-purification of Rec and Staufen-1 with APOBEC3G confirm the recent published subcellular co-localization studies that all three proteins are exist in stress granules. In addition, this is a further evidence for the connection between Rec associated RNA transport and HMM APOBEC3G granules. This hypothesis is confirmed by an experiment in this project because HERV-K(HML-2) RNA were detected in the HMM APOBEC3G granules. Furthermore it has been shown that with an additionally transfection of Rec, the presence of HERV-K(HML-2) RNA in the HMM APOBEC3G granules can be increased. Studies have shown for the first time that with an ectopic expression of APOBEC3G the HERV-K113 particles decline. This indicates that the HMM APOBEC3G restriction mechanism is also effective against human endogenous retroviruses. Since the viral RNA is bound in the HMM APOBEC3G granules it is no longer disposable for protein synthesis or particle formation. This leads to a reduction of formed virus particles and might be protective against re-infection and re-integration.
