dc.description.abstractDas humane Genom besteht zu etwa 8 % aus viralen Erbinformationen von Retrovi-ren. Die Viren haben im Laufe der Evolution die Vorfahren des Menschen fortwäh-rend infiziert und werden seither vertikal über die Keimbahnzellen vererbt. Durch Mu-tationen und Deletionen können sich jedoch keine infektiösen und replikationsfähi-gen Viren mehr bilden. Ein Vertreter der HERV-K(HML-2) Familie, das HERV-K113, ist in der Lage virale Partikel zu bilden, jedoch sind auch diese nicht replikationsfä-hig. Zudem besitzt HERV-K113 offene Leserahmen für die viralen Proteine. Es konn-te gezeigt werden, dass in Krankheiten wie Krebs, Autoimmunkrankheiten und neu-rologischen Störungen die Aktivität von humanen endogenen Retroviren erhöht ist und die endogenen Retroviren womöglich zur Entstehung dieser Krankheiten beitra-gen. Außerdem konnte gezeigt werden, dass das Interferon-induzierbare Protein (IFI)-16 auf verschiedene Viren eine hemmende Wirkung hat und somit als Restrikti-onsfaktor agiert. Die postinsertionalen Mutationen in der HERV-K113 Sequenz wurden von Mitarbei-tern der Arbeitsgruppe ausfindig gemacht und mittels Mutagenese revertiert. Dies ermöglicht es, dass HERV-K113 als Molekularklon in seiner vermutlich ursprüngli-chen Form beim Zeitpunkt der Infektion des Menschen untersuchen zu können. Im Rahmen dieser Arbeit soll die Wirkung von IFI-16 auf die Viruspartikelproduktion nach Transfektion von Molekularklonen untersucht werden. Als Indikator für die Vi-ruspartikelproduktion wird die Aktivität der Reversen Transkriptase herangezogen, die in Ab- oder Anwesenheit von IFI-16 bestimmt wird. In dieser Arbeit konnte gezeigt werden, dass die Viruspartikelproduktion von HERV-K113 durch IFI-16 stark gehemmt wird. Wurde im Molekularklon jedoch der U3-Bereich der LTR-Region oder die gesamte LTR-Region mit einem CMV Promotor ausgetauscht, konnten diese nicht mehr gehemmt werden. Dies deutet darauf hin, dass nicht nur die RU5-Region des Virusgenoms entscheidend für die hemmende Wirkung von IFI-16 ist, sondern auch der U3-Bereich und somit die gesamte LTR-Region am 5‘-Ende. Des Weiteren konnte gezeigt werden, dass die Kernlokali-sierungssequenz des IFI-16 Proteins essenziell für die Restriktion von HERV-K113 ist. Die Ergebnisse legen nahe, dass IFI-16 seine hemmende Wirkung auf die virale Partikelproduktion von HERV-K113 im Zellkern ausübt.ger
dc.description.abstractThe human genome consists of approximately 8 % viral genetic information. These viruses infected the humans throughout the history of the humans and are transmit-ted vertical by the germline cells ever since. Due to mutations and deletions infec-tious and replicating virus can no longer be formed. HERV-K113 is a member of the HERV-K(HML-2) family and is able to form viral particles but still these particles are not infectious. Furthermore HERV-K113 owns open reading frames for the viral pro-teins. It was shown that in several diseases like cancer, autoimmune diseases and neurological disorders human endogenous retroviruses are more active and that these retroviruses perhaps contribute to the development of these diseases. It was shown that the interferon-inducible protein (IFI)-16 has an inhibitory effect on some viruses and therefore acts as a restriction factor. The postinsertional mutations in the sequence of the HERV-K113 had been identi-fied and reverted by the workgroup. In this way it is possible to examine the HERV-K113 molecular clone in its presumably original form at the point of infecting the hu-mans. The aim of this work is to examine the effect of IFI-16 on the viral particalpro-duction after the transfection of molecular clones. As an indicator for viral particle production the activity of the reverse transcriptase should be used and measured in the absence or presence of the IFI-16 protein. The results show a strong inhibition of the production of HERV-K113 virus-like parti-cles in the presence of the IFI-16 protein compared to the absence of the protein. If the U3-region of the molecular clone was exchanged with a CMV promotor with the RU5-region still present, there was no inhibition of the viral replication through IFI-16 shown. Virus-like particles containing a CMV promotor instead of the entire LTR-region were not inhibited through the IFI-16 protein. These finding indicate that not only the RU5-region of the viral genome is responsible for the inhibitory effect of IFI-16 but the entire LTR-region consisting of the U3, R and U5 regions. In addition, it could be shown that the nuclear localization signal of the IFI-16 protein is essential for the restriction of HERV-K113. The results indicate that IFI-16 affects the viral rep-lication cycle of HERV-K113 in the nucleus.eng
